Novel Approaches for the Design of Vaccines against Infectious and Autoimmune Diseases

چکیده
The renaissance in vaccine research during the past decade led to several new approaches towards vaccine development. One is the evaluation of the potential of synthetic peptides corresponding to sequences of microbial proteins, comprising defined epitopes, to elicit antibodies with capacity to inactivate the respective pathogens and to protect against infection. Focusing on influenza, a novel approach – synthetic recombinant vaccines – was developed, in which the relevant epitopes are expressed by genetic engineering. Oligonucleotides coding for three influenza epitopes stimulating B cells, T helper cells and cytotoxic T lymphocytes (CTLs), respectively, were individually inserted into the flagellin gene of a Salmonella vaccine strain. The protective immunity, even against a lethal challenge, conferred by a vaccine, comprising a mixture of these three constructs, persisted for at least seven months and was effective against several strains of influenza. These results are indicative of the potential of such synthetic constructs as a long-range and broad-spectrum vaccines against influenza in other viruses, including HIV. A similar approach led also to protective immunization against the parasite Schistosoma mansoni..

Application of bacterial shuttle vectors in designing new vaccines against infectious diseases: brief report
Background: Today, several vaccines have been developed to prevent infectious diseases. The older first-generation vaccines may have many problems. In this regard, genetic engineering plays an important role using tools such as shuttle vectors to develop recombinant DNA vaccines that usually include plasmid constructed so that can propagate in two different host species. The present study revie...
متن کاملPotential Vaccines for Treating Crohn\'s Disease
Crohn's disease (CD) is an inflammatory disease of the gastrointestinal tract (GIT) tract and can affect several parts of the digestive system. There is a relationship between impaired mucosal barrier in the GIT of inflammatory bowel disease patients and the role of bacteria such as Mycobacterium avium paratuberculosis in CD. Apart from different therapeutic approaches for treating CD, developm...

Introduction: Despite considerable efforts to control AIDS pandemic, it is still one of the significant infectious concerns worldwide. The advance in medical research has led to the development of highly active antiretroviral therapy with a considerable effect to suppress the disease. However, an effective vaccine capable of eradication the HIV pandemic is not available yet. Failure to develop ...

Background: Enterohemorrhagic Escherichia coli (EHEC) O157:H7 is an infectious zoonotic pathogen causing human infections. These infections, in some cases, can lead to hemolytic uremic syndrome and its life-threatening complications and even death worldwide. The first intimate bacterial adhesion, intimin (I), with its own receptor translocated intimin receptor (Tir) and E. coli secreted protein...

Mosquito borne infectious diseases are among important group of diseases worldwide. Vaccination is available for some tropical mosquito-borne diseases, especially for Japa-nese encephalitis virus infection and yellow fever. There are also several attempts to develop new vaccines for the other mosquito-borne diseases such as malaria, dengue infection and West Nile virus infection. In this articl...
